Resumo
The horizontal transfer of antibiotic resistance genes is a primary event for the emergence and dissemination of antibiotic resistance, further spread through clonal bacterial selection. Although horizontal gene transfer can be investigated based on phenotypic properties of bacteria, other methodological approaches are required to infer about the genetic recombination events or about the horizontal versus vertical transfer of genes. In this seminar, it will be reported the use of polyphasic approaches, involving conjugation assays, whole bacterial genome sequencing, combined with plasmid characterization based on pulsed field gel electrophoresis, long read sequencing PacBio and optical DNA mappingto unveil horizontal gene transfer of antibiotic resistance. These tools were used to assess the effect of metals on resistance dissemination, the genetic recombination of plasmids during conjugation or the mode of transmission, horizontal or vertical, that explains the dissemination of a given resistance type in a hospital. This is valuable information to investigate antibiotic resistance evolution and therefore, to contribute to its combat.
